Encourage comments by asking direct questions of the viewers, not just saying "Please comment". Also answer every comment, this will encourage repeat comments and others to comment as well.
As @babyteeth4 says, you need what is commonly referred to as a call to action. It's a term often used primarily by people who already know what it means but it is rarely explained to the newbie. It is something in your video encouraging the people who watch to do complete some action whether to comment, subscribe, buy a product, check out another video, etc. In standard marketing and advertising, there is a general rule of 3s. Listen to ads on the radio or otherwise for example. You'll notice that in a good ad, the most important information they want to convey (business name, location, etc.) is always mentioned at least 3 times, even in a 30 second radio spot. This is a psychological trigger as it takes repetition for the brain to move to action where inaction is standard and 3 repetitions is the ideal that has been proven useful without overdoing it.
I'm not great at this myself. I only ask for comments twice in each video. Once at the beginning and once at the end. It does work though, by providing thought provoking questions or topics that actually get people interested. Controversial topics are also good for the initial generation of discussion but can devolve into flaming/trolling very quickly. It's also best not to have multiple different calls to action as the more divergent thinking that occurs, the less impact each has.
What won't work, is tossing up a random gameplay video with no topic of discussion and expecting unsolicited feedback. If there's nothing interesting to comment on, then there simply won't be any comments.
