Congratulations on getting so close to a wonderful milestone!
Here's what I think, play something that you can get heavily invested into.
I would say that a 50 subscriber special should be worthwhile, lengthy, and appropriate.
So play something that can be continued. Something heavy. Something substantial.

Ah, so what you are saying is play a story driven game for half an hour, then every subsequent episode be 15 minutes as standard? That could work well.
And Ponggave me an idea along with that, maybe one shorter game, and one long game to start off a new series.
Edit - To clarify, reached 50 a few days ago, but not had the time to do anything substantial, but have a good amount of time blocked off tonight to record
