Monetizing Question! Please help!

I'm quite new to monetizing videos and I'm trying new things. I know some of the basics but I'm really curious about something. I see that there is a box that you can check so that Advertisements will appear at that given time.

Example is posted below.

Can someone please tell me if you still generate more money for the advertisements that come up after the first one?

well... The AD's I'm using is only overlay ads because I want people to see my content over the ads! Hopefully that isn't a big problem.
