I use paint.net for all my thumbnails because its free plus the big plugin pack is kickass. Lets me do everything a lot easier then photoshop.

As for fonts go to: http://www.dafont.com/

It really depends on what kinda video. If it's a minecraft video I use a minecraft type font. If it's a comical type game like AVGN Adventures I use a font similar to something that fits his logo. Same with other stuff. Just depends really.

As per what is 'best', just make sure its clear and readable at a small size :)

What is the best font? Well you could use just a list of common fonts. It's really up to you, just make sure its readable, in your program zoom out a bit till the image is thumnail size. Can you read the font easily? There you go.

Just make sure to stay away from some of the more fancy decrorated fonts. Kinda keep it basic, like a sans-serif font. And also aim for bolder lettering, and a drop shadow or stroke.

I like chunkier fonts the most myself for thumbnails as most of them are easy to read, skinny fonts tend to blend too easily into the thumbnail imagery and its harder to make it stand out. I use dafont most of the time too.

I feel that the thumbnail font has to stand out and be eye catching as it is the first thing that your viewers would see.
